Epitalon (also spelled Epithalon or Epithalone) is a synthetic tetrapeptide (Ala-Glu-Asp-Gly) based on epithalamin, a polypeptide extract from the bovine pineal gland. It was developed by Professor Vladimir Khavinson at the Saint Petersburg Institute of Bioregulation and Gerontology, where decades of research have established it as the most direct pharmacological activator of telomerase — the enzyme responsible for maintaining telomere length. Telomeres are the protective nucleotide sequences (TTAGGG repeats) at the ends of chromosomes that shorten with each cell division. When telomeres reach a critical minimum length, cells enter replicative senescence. Epitalon's ability to reactivate telomerase positions it at the center of cellular aging research.
Epitalon activates the expression of the human telomerase reverse transcriptase (hTERT) gene, the catalytic subunit of telomerase. Telomerase adds TTAGGG nucleotide repeats to the 3' ends of telomeric DNA, counteracting the progressive shortening that occurs during DNA replication. Research by Khavinson's group has demonstrated that Epitalon treatment increases telomere length in human cell cultures, extends the replicative lifespan of somatic cells beyond the Hayflick limit, and reactivates telomerase in cells where it has become silenced. Beyond telomerase activation, Epitalon stimulates melatonin production by the pineal gland, restoring the age-related decline in nocturnal melatonin secretion that contributes to circadian rhythm disruption, sleep disturbance, and reduced antioxidant protection.
The longevity research on Epitalon is extensive within the Russian biogerontology literature. Studies in animal models demonstrate extended mean and maximum lifespan (15-25% increases in some cohorts), reduced incidence of spontaneous tumors, improved immune function, and normalized neuroendocrine regulation. Human observational studies in elderly populations show improved physical endurance, sleep quality, immune markers, and melatonin rhythms. While the peer-reviewed English-language literature on Epitalon is growing, the Khavinson group's body of work spanning three decades represents the most comprehensive dataset on any telomerase-activating compound.
Epitalon is suited for researchers investigating telomere biology, replicative senescence, pineal gland function, circadian rhythm restoration, and systemic aging. It is the most accessible telomerase-activating compound available and a foundational element of longevity research protocols.
Reconstitute with 1-2ml bacteriostatic water. Administer via subcutaneous injection, typically 5-10mg per day, in protocols of 10-20 consecutive days repeated every 4-6 months. This cyclical dosing pattern follows the original clinical research protocol and reflects the observation that telomerase activation triggers a sustained downstream effect that persists beyond the dosing window. Some researchers prefer intranasal administration for convenience. Store at 2-8C after reconstitution. Lyophilized powder is stable at room temperature but refrigeration is recommended. The 10mg vial supports 1-2 dosing cycles at standard research parameters.
Epitalon is supplied as a lyophilized (freeze-dried) powder and must be reconstituted with bacteriostatic water (BAC water) before use in a research setting.
- Clean the BAC water vial stopper and the peptide vial stopper with an alcohol swab. Allow to dry.
- Draw the required volume of BAC water into a sterile syringe (typically 1–3 mL depending on target concentration).
- Angle the needle so the water runs down the inside wall of the peptide vial. Avoid dispensing directly onto the powder.
- Do not shake. Gently swirl or roll until fully dissolved. Vigorous shaking can denature peptides.
- Refrigerate reconstituted solution at 2–8°C. Most reconstituted peptides are stable 14–30 days depending on compound.
