CJC-1295 without DAC, also known as Modified GRF (1-29) or Mod-GRF, is a synthetic analog of the first 29 amino acids of human growth hormone-releasing hormone (GHRH). The native GHRH(1-29) fragment, also called Sermorelin, has a plasma half-life of less than 10 minutes due to rapid enzymatic degradation. CJC-1295 No DAC incorporates four amino acid substitutions at positions 2, 8, 15, and 27 that protect it from DPP-4 and other proteases, extending its effective half-life to approximately 30 minutes. This modification preserves the natural pulsatile pattern of GH release while providing meaningful protection against degradation.

As a GHRH analog, CJC-1295 No DAC binds to the GHRH receptor on anterior pituitary somatotroph cells, activating the adenylyl cyclase-cAMP-PKA signaling cascade. This triggers calcium influx through voltage-gated calcium channels, which stimulates the exocytosis of stored growth hormone vesicles. Importantly, CJC-1295 No DAC amplifies the body's natural GH pulses rather than creating a sustained artificial elevation. This preserves the physiological pulsatile secretion pattern that is critical for GH receptor sensitivity — continuous GH exposure (as seen with exogenous HGH) leads to receptor downregulation, while pulsatile release maintains receptor responsiveness.

Research has documented that CJC-1295 No DAC produces significant increases in GH amplitude (peak GH levels during pulse) without substantially altering pulse frequency. Studies show 2-10 fold increases in GH secretion following administration, with concurrent elevations in IGF-1 of 40-100% above baseline. The preservation of natural pulsatility makes it particularly attractive to researchers investigating GH optimization without the side-effect profile of exogenous HGH administration. CJC-1295 No DAC is best suited for researchers investigating natural-pattern GH optimization, anti-aging protocols, recovery enhancement, and body composition improvement. It is almost always used in combination with a GHRP (growth hormone releasing peptide) such as Ipamorelin — the CJC/Ipa combination is the most widely referenced GH peptide stack in the research community. The GHRH analog (CJC) provides the release signal while the GHRP (Ipamorelin) amplifies it, producing a synergistic GH pulse 3-5 times greater than either compound alone.

Reconstitute with 2.5ml bacteriostatic water (yielding 2mg/ml). Administer via subcutaneous injection, typically 100mcg per dose, 1-3 times daily. Optimal timing is pre-sleep (when natural GH pulse is strongest), upon waking, and/or post-workout. The 30-minute half-life means timing matters — inject 15-30 minutes before sleep or food. Store at 2-8C after reconstitution. CJC-1295 No DAC is sensitive to heat and agitation; avoid shaking the vial.

CJC-1295 No DAC is supplied as a lyophilized (freeze-dried) powder and must be reconstituted with bacteriostatic water (BAC water) before use in a research setting.

  1. Clean the BAC water vial stopper and the peptide vial stopper with an alcohol swab. Allow to dry.
  2. Draw the required volume of BAC water into a sterile syringe (typically 1–3 mL depending on target concentration).
  3. Angle the needle so the water runs down the inside wall of the peptide vial. Avoid dispensing directly onto the powder.
  4. Do not shake. Gently swirl or roll until fully dissolved. Vigorous shaking can denature peptides.
  5. Refrigerate reconstituted solution at 2–8°C. Most reconstituted peptides are stable 14–30 days depending on compound.
